The Perlin Lab at the University of Louisville is seeking a Lab Technician to join their research team focused on plant/parasite interactions, particularly involving smut fungi.
Key Responsibilities:
Cultivate wildflower species and infect them with fungi to study host/pathogen interactions.
Extract and purify RNA from fungi, plants, and infected plants.
Produce complementary cDNA and perform real-time qPCR.
Utilize a Nanodrop spectrophotometer for nucleic acid quantification.
Prepare and sterilize growth media, buffers, and solutions using autoclaving techniques.
Handle and maintain laboratory equipment, ensuring proper sterilization and functionality.
Conduct standard molecular biology techniques, including plasmid DNA isolation, agarose gel electrophoresis, polymerase chain reaction (PCR), DNA fragment purification, and Gibson Assembly for cloning.
Perform BLAST analysis of DNA and protein sequences.
Preferred Qualifications:
Completion of coursework in Genetics; courses in Cell Biology and Microbiology are preferred.
Proficiency in sterile technique and proper handling of microbial cultures.
Extensive experience in growing wildflower species and infecting them with fungi.
Hands-on experience with RNA extraction and purification, cDNA synthesis, and qPCR.
Familiarity with laboratory equipment such as the Nanodrop spectrophotometer.
Ability to prepare and sterilize growth media, buffers, and solutions.
Experience with standard molecular biology methods, including plasmid DNA isolation, agarose gel electrophoresis, PCR, DNA fragment purification, and Gibson Assembly.
